of macrophages have been described in the human liver, their function and turnover in patients with obesity at high risk of developing non-alcoholic fatty liver disease (NAFLD) and cirrhosis are currently unknown. Herein, we identify a specific human population of resident liver myeloid cells that protects against the metabolic impairment associated with obesity. By studying the turnover of liver myeloid cells in individuals undergoing liver transplantation, we find that liver myeloid cell turnover differs between humans and mice. Using single-cell techniques and flow cytometry, we determine that the proportion of the protective resident liver myeloid cells, denoted liver myeloid cells 2 (LM2), decreases during obesity. Functional validation approaches using human 2D and 3D cultures reveal that the presence of LM2 ameliorates the oxidative stress associated with obese conditions. Our study indicates that resident myeloid cells could be a therapeutic target to decrease the oxidative stress associated with NAFLD.", "doi": "10.1038/s42255-023-00834-7", "pmid": "37414931", "labels": {"Spatial Proteomics": "Service"}, "xrefs": [{"db": "pmc", "key": "PMC10365994"}, {"db": "pii", "key": "10.1038/s42255-023-00834-7"}], "notes": [], "created": "2023-11-21T15:12:13.445Z", "modified": "2023-11-21T15:12:13.896Z"}, {"entity": "publication", "iuid": "17cbfb54c2314acab08b8b2221e6e82f", "links": {"self": {"href": "https://publications.scilifelab.se/publication/17cbfb54c2314acab08b8b2221e6e82f.json"}, "display": {"href": "https://publications.scilifelab.se/publication/17cbfb54c2314acab08b8b2221e6e82f"}}, "title": "Impaired phosphocreatine metabolism in white adipocytes promotes inflammation.", "authors": [{"family": "Maqdasy", "given": "Salwan", "initials": "S", "orcid": "0000-0001-5164-9879", "researcher": {"href": "https://publications.scilifelab.se/researcher/809a82e9fa1e4173ba1dd2133a91d5c5.json"}}, {"family": "Lecoutre", "given": "Simon", "initials": "S", "orcid": "0000-0002-2536-6119", "researcher": {"href": "https://publications.scilifelab.se/researcher/ac1619986a25402b994efa1811b5cfff.json"}}, {"family": "Renzi", "given": "Gianluca", "initials": "G"}, {"family": "Frendo-Cumbo", "given": "Scott", "initials": "S", "orcid": "0000-0002-9047-4650", "researcher": {"href": "https://publications.scilifelab.se/researcher/4bfa1b27416e47fd90a77657fcb0a34e.json"}}, {"family": "Rizo-Roca", "given": "David", "initials": "D"}, {"family": "Moritz", "given": "Thomas", "initials": "T", "orcid": "0000-0002-4258-3190", "researcher": {"href": "https://publications.scilifelab.se/researcher/95ad5b7fe48f42eda1328f54a385e097.json"}}, {"family": "Juvany", "given": "Marta", "initials": "M"}, {"family": "Hodek", "given": "Ondrej", "initials": "O"}, {"family": "Gao", "given": "Hui", "initials": "H", "orcid": "0000-0002-7306-7471", "researcher": {"href": "https://publications.scilifelab.se/researcher/c212c77475f042e284e0cbe9ae4c38fa.json"}}, {"family": "Couchet", "given": "Morgane", "initials": "M"}, {"family": "Witting", "given": "Michael", "initials": "M", "orcid": "0000-0002-1462-4426", "researcher": {"href": "https://publications.scilifelab.se/researcher/b522a40ff2e84e76a4e6d83fa1e8ff0a.json"}}, {"family": "Kerr", "given": "Alastair", "initials": "A", "orcid": "0000-0003-2085-1542", "researcher": {"href": "https://publications.scilifelab.se/researcher/076ba1b03b78451ba1ddb95b04e74b27.json"}}, {"family": "Bergo", "given": "Martin O", "initials": "MO"}, {"family": "Choudhury", "given": "Robin P", "initials": "RP"}, {"family": "Aouadi", "given": "Myriam", "initials": "M", "orcid": "0000-0001-6256-7107", "researcher": {"href": "https://publications.scilifelab.se/researcher/7ea7bb2be61a4e818685466d1005659d.json"}}, {"family": "Zierath", "given": "Juleen R", "initials": "JR", "orcid": "0000-0001-6891-7497", "researcher": {"href": "https://publications.scilifelab.se/researcher/f4ef2b94ba2d44dd9b41192567a7af53.json"}}, {"family": "Krook", "given": "Anna", "initials": "A", "orcid": "0000-0002-0891-0258", "researcher": {"href": "https://publications.scilifelab.se/researcher/f43339bcd571486b845a3b10e241bcc4.json"}}, {"family": "Mejhert", "given": "Niklas", "initials": "N", "orcid": "0000-0003-1785-833X", "researcher": {"href": "https://publications.scilifelab.se/researcher/81eb60b84ea14ad9ba7e4d3433c37a3e.json"}}, {"family": "Ryd\u00e9n", "given": "Mikael", "initials": "M", "orcid": "0000-0003-4785-1876", "researcher": {"href": "https://publications.scilifelab.se/researcher/ab61ac66c36749019ce5066fb1a5061e.json"}}], "type": "journal article", "published": "2022-02-00", "journal": {"title": "Nat Metab", "issn": "2522-5812", "volume": "4", "issue": "2", "pages": "190-202", "issn-l": "2522-5812"}, "abstract": "The mechanisms promoting disturbed white adipocyte function in obesity remain largely unclear. Herein, we integrate white adipose tissue (WAT) metabolomic and transcriptomic data from clinical cohorts and find that the WAT phosphocreatine/creatine ratio is increased and creatine kinase-B expression and activity is decreased in the obese state. In human in vitro and murine in vivo models, we demonstrate that decreased phosphocreatine metabolism in white adipocytes alters adenosine monophosphate-activated protein kinase activity via effects on adenosine triphosphate/adenosine diphosphate levels, independently of WAT beigeing. This disturbance promotes a pro-inflammatory profile characterized, in part, by increased chemokine (C-C motif) ligand 2 (CCL2) production. These data suggest that the phosphocreatine/creatine system links cellular energy shuttling with pro-inflammatory responses in human and murine white adipocytes. Our findings provide unexpected perspectives on the mechanisms driving WAT inflammation in obesity and may present avenues to target adipocyte dysfunction.", "doi": "10.1038/s42255-022-00525-9", "pmid": "35165448", "labels": {"Swedish Metabolomics Centre": "Service"}, "xrefs": [{"db": "pmc", "key": "PMC8885409"}, {"db": "pii", "key": "10.1038/s42255-022-00525-9"}], "notes": [], "created": "2022-12-05T08:10:40.997Z", "modified": "2025-10-17T13:03:15.360Z"}]}
